Research: Clustered Regularly Interspaced Short Palindromic Repeats (CRISPRs) are small RNAs that, together with CRISPR-associated (cas) genes, provide adaptive immunity in prokaryotes. CRISPR loci are present in half of all sequenced bacterial genomes and comprise several unique short spacer sequences, which are interspaced by conserved direct repeats. Spacers are acquired from foreign nucleic acids, (eg. invading bacteriophage), such that CRISPRs provide a remarkably dynamic adaptive immune system in both bacteria and archaea.
I investigate the function, diversity and evolution of CRISPRs in Salmonella, the #1 bacterial cause of food-borne illness in the USA. In collaboration with the PA Dept. of Health, Centers for Disease Control and Prevention and US Food and Drug Administration, I have established a rapid and accurate molecular subtyping method, CRISPR-MVLST, which uses CRISPR sequence information to separate Salmonella subtypes.
